Unit 1 · Electrochemical Methods of Analysis 10 hrs
Potentiometry — electrode potential and the electrochemical cell, the construction and working of reference, indicator and membrane electrodes, measurement of potential and pH, potentiometric titrations and end-point detection, and Karl Fischer titration; conductometry — the conductivity cell, conductometric titrations and applications; and polarography — residual, migration, diffusion and limiting currents, the dropping mercury electrode, the polarographic wave, the Ilkovic equation and applications.
Unit 2 · UV-Visible and Infrared Spectroscopy 10 hrs
The fundamentals of spectroscopy — the properties of electromagnetic radiation and the electromagnetic spectrum; UV-visible spectroscopy — the Beer–Lambert law and its deviations, electronic transitions, chromophores and auxochromes, spectral shifts and solvent effects, instrumentation (sources, wavelength selectors, sample cells and detectors) and single- and multi-component pharmaceutical analysis; and IR spectroscopy — fundamental vibrational modes, factors affecting vibrations, dispersive and FTIR instrumentation with sample-handling techniques, and pharmaceutical applications.
Unit 3 · Fluorimetry 7 hrs
The theory of fluorescence and phosphorescence; the factors affecting fluorescence including quenching; instrumentation of the spectrofluorimeter; and the pharmaceutical applications of fluorimetric analysis.
Unit 4 · Chromatographic Techniques and Electrophoresis 8 hrs
An introduction to chromatographic separation and its underlying principles; gel-filtration and affinity chromatography and their applications; and electrophoresis — the factors affecting electrophoretic mobility and the techniques of paper, gel and capillary electrophoresis with their applications.
Unit 5 · Instrumental Chromatography 10 hrs
Gas chromatography — theory, instrumentation, derivatization, temperature programming, advantages, disadvantages and applications; high-performance liquid chromatography — theory, instrumentation, advantages and applications; and HPTLC — principle, instrumentation and applications.
